Plasma were obtained from blood samples to which heparin (25 \u00b5g), were added to 10 ml tube before centrifugation at 900 g.,\u00a0 plasma (0.5\u00a0ml) in ground-glass centrifuge tube, was added to 0.2<sup>\u00a0<\/sup>ml of 20% perchloric acid, mixed well, and centrifuged at 850<sup>\u00a0<\/sup><em>g<\/em> for 10\u00a0min at 4\u00b0C. The supernatant was removed and filtered through a 22-\u00b5m Millipore GV<sub>13<\/sub> syringe filter. A 20-\u00b5l aliquot of the filtrate was analyzed by HPLC.<\/p>\n<p><strong>Assay by HPLC<\/strong><\/p>\n<p>High performance liquid chromatography (HPLC) 10AVP, Shimadzu, Japan, was used to determine the concentration of allopurinol in plasma.\u00a0 Plasma (0.5\u00a0ml) was added to 0.2<sup>\u00a0<\/sup>ml of 20% perchloric acid, mixed well, and centrifuged at 850<sup>\u00a0<\/sup><em>g<\/em> for 10\u00a0min at 4\u00b0C. The supernatant was removed and filtered through a 22-\u00b5m Millipore GV<sub>13<\/sub> syringe filter.\u00a0 A 20-\u00b5l aliquot of the filtrate was analyzed by using a Shimadzu 10AVP,\u00a0HPLC machine, operating in isocratic mode, equipped with a suplico \u00b5Bondapak C<sub>18<\/sub> column (4.6 mm i.d. x 250 mm, 5 \u00b5m particle size), an absorbance detector set at 254\u00a0nm. The mobile phase was 50\u00a0mM KH<sub>2<\/sub>PO<sub>4<\/sub>, pH 6.0,\u00a0at a flow rate of 1\u00a0ml\/min. Authentic allopurinol standards were run under similar conditions, and these values were used to calculate concentrations of allopurinol in plasma samples.<\/p>\n<p><strong>Linearity of the Chromatographic Method<\/strong><strong>\u00a0<\/strong><\/p>\n<p>The typical chromatogram obtained for a standard of allopurinol is shown in Fig. 1. The coefficient of variation was 1.12 %. Injection precision less than 5% RSD (Relative Standard Deviation) are considered appropriate for these trace level determinations. The accuracy of the method was estimated by injecting five different standard of the same concentration (1 mcg\/ml) obtaining a recovery of 99.6% with an RSD of 1.6%.<\/p>\n<p><strong>Recovery of the Method<\/strong><\/p>\n<p>Table 2 show the results obtained in the analysis of the extracts from spiked plasma with different quantities of allopurinol (0.78 mcg. 10.2 mcg and 26.7 mcg); the average recovery was 98.5%.\u00a0\u00a0 The RSD values ranged from 1.6% for a quantity of 10.2 \u03bcg (intra-day) to 5.6% for 0.78 \u03bcg in the inter-day study.<\/p>\n<p><strong>Statistical Analysis<\/strong><\/p>\n<p>The pharmacokinetic characteristics of allopurinol were determined from the plasma concentration-time data. Peak plasma concentration (C<sub>max<\/sub>) and time to maximum plasma concentration (T<sub>max<\/sub>) were determined directly from raw data. The area under the curve (AUC<sub>0-t<\/sub>, from 0 to last measured concentration) was calculated by the trapezoidal method.<\/p>\n<p>Student\u2019s (t)-test was used to estimate the significance of Table2\u00a0display\u00a0the\u00a0effects\u00a0obtained\u00a0within the\u00a0analysis\u00a0of the extracts from spiked plasma with different\u00a0portions\u00a0of allopurinol (0.78\u00a0mcg. 10.2 mcg and 26.7 mcg); the average\u00a0restoration\u00a0turned into\u00a098.5%. The RSD values ranged from 1.6% for a\u00a0quantity\u00a0of 10.2 \u03bcg (intra-day) to\u00a05.6 % for\u00a00.78\u00a0\u03bcg\u00a0within the\u00a0inter-day study.<\/p>\n<p>Plasma concentrations of bioavailability of both formulations judged by the calculated pharmacokinetic parameters listed in Tables 5 and 6 and Figures 3 and 4.\u00a0\u00a0 Among the parameters utilized for comparison between the two formulations were the area under curve (AUC) calculated from the plasma concentration profile (Fig. 5).\u00a0\u00a0 The area was estimated by combination of linear \u2013log \u2013 linear trapezoidal method which yields more acceptable results than the linear method alone (23).<\/p>\n<p>The\u00a0most\u00a0concentrations of allopurinol for\u00a0each\u00a0300mg hyporic and 300mg zyloric\u00a0were\u00a029.8 and 30.6mg at\u00a0maximum\u00a0time of 1.5\u00a0hours for\u00a0both\u00a0formulation.
